Wister大鼠每只每周口饲AFB_1 60μg,35周后增加15μg,63周后停毒,总量为4.10mg/只。实验开始后,每月处死一次、自45周起改为半个月一次。肝组织及肝癌结节作扫描和透射电镜观察。AFB_1对肝细胞超微结构方面的影响主要表现在: 诱癌早期(1~7周):6周前,肝细胞无明显改变,自7周起出现少量早期增生性肝细胞。在胞质中有大量的板层状或簇状分布的粗面内质网,线粒体数量不定,基质变深,脊变异,糖原减少,高不基器不常见。未见细胞核有明显改变。细胞膜变化不显著。
The Wister rats were fed with 60 μg AFB_1 per week, 15 μg 35 weeks later and 63 weeks later with a total amount of 4.10 mg. After the experiment began, it was sacrificed once a month for half a month from 45 weeks onwards. Scanning and transmission electron microscopic observation of liver and liver cancer nodules. The effects of AFB_1 on the ultrastructure of hepatocytes were mainly manifested in the early stage of inducing cancer (1-7 weeks): no significant change of hepatocytes was observed 6 weeks ago, and few early proliferative hepatocytes appeared from the 7th week. In the cytoplasm of a large number of lamellar or cluster-like distribution of rough endoplasmic reticulum, mitochondrial variable number of matrix darker, spinal variability, glycogen reduction, high substrate is not uncommon. No significant change in the nucleus was observed. Cell membrane changes are not significant.
