目的建立检测氟喹诺酮类药物的免疫亲和层析样品前处理方法结合高效液相色谱法的分析方法。方法通过将抗恩诺沙星单抗与溴化氰活化的琼脂糖4B偶联作为免疫吸附剂来提取胎牛血清中的5种氟喹诺酮类药物,并用高效液相色谱仪配备荧光检测器检测。结果在本研究中,诺氟沙星、恩诺沙星、环丙沙星、沙拉沙星和达氟沙星在胎牛血清中的萃取回收率分别为82.97%、70.50%、70.16%、31.83%和6.37%,检出限均为0.5 ng/m L。结论建立的氟喹诺酮类免疫亲和层析色谱胶具有高的吸附容量、选择性、萃取效率和稳定性。免疫亲和色谱法是一种可以从复杂基质样品中萃取多种氟喹诺酮抗生素的可行方法。
Objective To establish a method for the analysis of fluoroquinolones by immunoaffinity chromatography and high performance liquid chromatography (HPLC). Methods Five fluoroquinolones in fetal bovine serum were extracted by conjugating anti-enrofloxacin monoclonal antibody and cyanogen bromide-activated agarose 4B, and detected by high performance liquid chromatography with a fluorescence detector . Results In this study, the extraction recoveries of norfloxacin, enrofloxacin, ciprofloxacin, sarafloxacin and danofloxacin in fetal bovine serum were 82.97%, 70.50%, 70.16%, 31.83 % And 6.37% respectively. The limits of detection were 0.5 ng / m L. Conclusion The established fluoroquinolone immunoaffinity chromatography chromatography gel has high adsorption capacity, selectivity, extraction efficiency and stability. Immunoaffinity chromatography is a viable method for extracting multiple fluoroquinolone antibiotics from complex matrix samples.
