本文报道了脑血流显象剂六甲基丙二胺肟(HM-PAO)的合成、异构体的分离及标记。将~(99m)Tc标记的d1-、meso-和mixture-HM-PAO从层析、体内分布及家兔显象等方面进行了比较。结果显示:~(99m)Tc-dl-HM-PAO以丁酮作展开剂的硅胶G薄板层析中的Rf值(0.8~1.0)大于~(99)mTc-meso-HM-PAO(0.6~0.8),且dl异构体较meso—HM-PAO和mixture-HM-PAO在脑内浓聚更高并更稳定,与英国Amersham公司所制dl-HM-PAO药盒Ceretec~(TM)完全一致。临床初步应用获得满意结果。
This paper reports the synthesis of cerebral blood flow imaging agent hexamethylpropylenediamine oxime (HM-PAO), the separation and labeling of isomers. The ~ (99m) Tc-labeled d1-, meso- and mixture-HM-PAO were compared in terms of chromatography, in vivo distribution, and rabbit visualization. The results showed that the Rf value (0.8 ~ 1.0) of ~ (99m) Tc-dl-HM-PAO using methyl ethyl ketone as developing solvent was higher than that of 99mTc-meso-HM- 0.8), and the dl isomer was more concentrated and more stable in the brain than the meso-HM-PAO and mixture-HM-PAO. The dl-HM-PAO kit Ceretec ™ Consistent. Preliminary clinical application to obtain satisfactory results.
