The aim of the study was to find out the mutation frequency spectrum for determining the specific codon and mutation type of P53 gene in ovarian cancer. Also detected P53 gene mutation in the tumor, P53 allele deletion rate. Tissue samples were taken from 63 surgically excised ovarian cancers that were histologically examined to determine that the sample contained more than 75% of the tumor cells. Total RNA was extracted from cancer specimens and paired non-cancerous tissue areas or leukocytes, respectively. Purified RNA was reverse transcribed to generate a cDNA copy of exon 4-10 of P53 gene. The experiment and control group extracts were respectively subjected to PCR reaction to generate sufficient templates for DNA sequence analysis.
