目的建立测定菟丝子中绿原酸、金丝桃苷、异槲皮苷、紫云英苷、山奈酚的含量方法。方法采用高效液相色谱(HPLC)法。色谱柱为Zorbax SB C18;流动相为乙腈-0.1%磷酸溶液,梯度洗脱;绿原酸的检测波长为327 nm,金丝桃苷、异槲皮苷、紫云英苷、山奈酚的检测波长为360 nm;流速为1.0 mL·min-1;柱温为30℃。结果 5种活性成分均达到基线分离,线性关系良好(r≥0.9998);平均加样回收率为96.8%~101.0%,RSD<3.0%(n=6)。结论该方法灵敏,可靠,可用于菟丝子药材的质量评价。
Objective To establish a method for the determination of chlorogenic acid, hyperoside, isoquercitrin, albinoside and kaempferol in dodder. Methods High performance liquid chromatography (HPLC) method was used. The column was Zorbax SB C18; the mobile phase consisted of acetonitrile-0.1% phosphoric acid solution with gradient elution; the detection wavelength of chlorogenic acid was 327 nm. The detection of hyperoside, isoquercitrin, The wavelength was 360 nm; the flow rate was 1.0 mL · min-1; the column temperature was 30 ℃. Results The five active ingredients all achieved baseline separation with good linearity (r ≥ 0.998). The average recovery was 96.8% -101.0% with RSD <3.0% (n = 6). Conclusion The method is sensitive and reliable and can be used to evaluate the quality of Dodder.
