为观察白介素1(IL-1)和白介素1受体拮抗剂(IL-1Ra)在人发育骨与软骨组织中的表达及分布,认识IL-1系统在人发育骨与软骨中的作用,用组织学及免疫组化技术,对临床经过石蜡包埋的人发育关节软骨组织进行定位定性检测,结果显示3月龄胚胎的软骨组织,IL-1β在肥大样改变的软骨细胞染色阳性。在4~6月龄胎儿,生长板软骨细胞自增殖后期到肥大样改变,IL-1β染色阳性。骨骺软骨(未钙化)内新生髓腔周围软骨细胞及骨髓腔内成骨细胞IL-1β也呈阳性染色。IL-1Ra蛋白表达区域基本类似于IL-1β,但表达的量较多,区域分布较广泛。结果显示,IL-1和IL-1Ra可能在人发育骨与软骨组织中通过旁分泌及自分泌方式协同发挥作用。
To observe the expression and distribution of interleukin-1 (IL-1) and interleukin-1 receptor antagonist (IL-1Ra) in human bone and cartilage, to understand the role of IL-1 in human development of bone and cartilage, Histological and immunohistochemical techniques were used to characterize the clinical development of articular cartilage in paraffin-embedded human. The results showed that the cartilage tissue and IL-1β in 3-month-old embryos were stained positively in the hypertrophic-like chondrocytes. In 4- to 6-month-old fetuses, growth plate chondrocytes changed from hypertrophy to hypertrophy, and IL-1β staining was positive. Epiphyseal cartilage (uncalcified) around the new bone marrow cavity and intramedullary osteoblast IL-1β also positive staining. IL-1Ra protein expression region is basically similar to IL-1β, but the amount of expression is more, the region is more widely distributed. The results show that IL-1 and IL-1Ra may play a synergistic role in paracrine and autocrine modes in human bone and cartilage.
