目的:建立硫堇瑞利共振散射荧光法测定肝素钠的含量。方法:在近中性的Britton Robinson(B-R)缓冲溶液中,肝素钠与硫堇形成离子缔合物,在564 nm处生成新的瑞利共振散射(RRS)峰,随着肝素钠浓度的增加RRS峰显著增强。结果:在0.025~0.500 mg·L-1范围内RRS强度与肝素钠浓度呈良好的线性关系,线性回归方程为ΔI=0.125+16.88C,平行6次测定,RSD为1.67%~3.01%,方法回收率为104.0%~106.4%。结论:硫堇染料作为增色剂的瑞利共振散射荧光法用于肝素钠注射液含量测定结果满意。
PURPOSE: To establish a method for the determination of heparin sodium by resonance fluorescence scattering assay. METHODS: In a nearly neutral Britton Robinson (BR) buffer solution, sodium heparin and thionine formed an ion association complex, generating a new Rayleigh resonance scattering (RRS) peak at 564 nm. As the heparin concentration increased RRS peak increased significantly. Results: There was a good linear relationship between the concentration of heparin and the concentration of heparin in the range of 0.025-0.500 mg · L-1, the linear regression equation was ΔI = 0.125 + 16.88C, and the RSD was 1.67% ~ 3.01% The recovery rate was 104.0% ~ 106.4%. Conclusion: Thionine dye as a toner enhancer by Rayleigh resonance scattering fluorescence method for the determination of heparin sodium injection results were satisfactory.
