目的 :制备含胞嘧啶转氨酶 (CD)基因的重组腺病毒 (Ad- CD)并进行体外肿瘤的自杀基因治疗。方法 :构建含 CD或L ac Z基因的重组腺病毒载体 ,在 2 93细胞内重组包装后 ,体外感染小鼠结肠癌细胞株 C2 6 ,并给予不同浓度的前药氟胞嘧啶(5 - FC)进行肿瘤杀伤效果观察。 结果 :重组腺病毒载体构建成功。用制备的 Ad- CD液感染 C2 6细胞 ,并予以不同浓度 5 - FC后 ,肿瘤细胞生长受不同程度抑制 ,而对照组细胞和原位转 L ac Z基因的 C2 6细胞的生长未出现明显的变化。结论 :应用腺病毒载体转 CD基因进行体外肿瘤治疗效果明显 ,为自杀基因应用于体内结肠癌的基因治疗研究奠定基础
Objective: To prepare recombinant adenovirus (Ad-CD) containing cytosine aminotransferase (CD) gene and perform suicide gene therapy on tumor in vitro. METHODS: A recombinant adenoviral vector containing the CD or Lac Z gene was constructed. After recombinant packaging in 293 cells, the mouse colon carcinoma cell line C26 was infected in vitro and given different concentrations of the prodrug flucytosine (5-FC). ) Observation of tumor killing effect. Results: The recombinant adenovirus vector was successfully constructed. After the C2 6 cells were infected with the prepared Ad-CD fluid and different concentrations of 5-FC were added, the growth of tumor cells was inhibited to different extents, but the growth of the control cells and the C2 6 cells transformed to the Lac Z gene in situ was not obvious. The change. Conclusion : The effect of adenovirus vector transfection of CD gene on tumor therapy in vitro is obvious. It lays a foundation for gene therapy of suicide gene in colon cancer in vivo.
