用二亚硝基哌嗪(DNP)1μg/ml和10μg/ml处理人胚鼻咽成纤维细胞,与空白对照细胞比较:均见细胞有形态变化,细胞呈异形性和核浆比例增大;细胞倍增时间缩短,饱和密度增加,在一定程度上失去接触抑制,在低血清(1%)条件下能维持生长和传代;染色体变化出现较早,二倍体随代数增加而减少,而高异倍体则增加,染色体结构出现断裂、断片、双微体、双着丝粒染色体和环状染色体等畸变;细胞能在双层半固体软琼脂上生长;细胞寿命延长。
Human embryonic nasopharyngeal fibroblasts were treated with dinitrosopiperazine (DNP) 1μg/ml and 10μg/ml. Compared with blank control cells, there were morphological changes in cells, heteromorphism of cells and increase in the proportion of nuclear plasma. Cell doubling time was shortened, saturation density was increased, contact inhibition was lost to a certain extent, and growth and passage were maintained under low serum (1%) conditions; chromosome changes occurred earlier, and diploids decreased with increasing generations, while high heterogeneity Phenoplasty increased, chromosome structure appeared broken, fragments, double microsomes, dicentric chromosomes and ring chromosome aberrations; cells can grow on double-layer semi-solid soft agar; cell life is prolonged.
