目的建立衍生化GC-MS同时测定不同来源鱼腥草药材中9种脂肪酸甲酯化衍生物的含量。方法采用Inowax(0.32mm×30m,0.25μm)弹性石英毛细管色谱柱,程序升温,初温:110℃保持3min,以2℃·min-1升温至164℃,保持10min,再以8℃·min-1升温至210℃,保持10min。进样口温度为230℃,分流比为20∶1,进样量1μL,柱流速2.5mL·min-1,对不同来源的鱼腥草药材石油醚提取物的甲酯化样品采用SIM方式进行分析。结果月桂酸、肉豆蔻酸、棕榈酸、棕榈油酸、硬脂酸、油酸、α-亚麻酸、亚油酸、γ-亚油酸等9种脂肪酸均获得良好线性,并测定其在不同来源的鱼腥草药材中含量。结论方法准确、专属,能有效测定鱼腥草药材中含有的脂肪酸。
Objective To establish a derivatized GC-MS for the simultaneous determination of 9 fatty acid methyl ester derivatives in fishes from different sources. Methods Inowax (0.32mm × 30m, 0.25μm) elastic quartz capillary column was used to increase the temperature. The initial temperature was kept at 110°C for 3min, and the temperature was raised from 2°C·min-1 to 164°C for 10min, then 8°Cmin. -1 Raise the temperature to 210 °C and hold for 10 min. The injection port temperature was 230°C, the split ratio was 20:1, the injection volume was 1 μL, and the column flow rate was 2.5 mL·min-1. Results The lauric acid, myristic acid, palmitic acid, palmitoleic acid, stearic acid, oleic acid, α-linolenic acid, linoleic acid, γ-linoleic acid and other 9 fatty acids all obtained good linearity and were determined to be different. The source of the content of fishy herbs in herbs. Conclusion The method is accurate and exclusive, and can effectively determine the fatty acids contained in the herbal medicines from the fish.
