目的 通过检测FⅧ相关抗原(von Willebrand factor,vWF)和层粘蛋白(laminin)在大鼠全脑缺血再灌注后脑微血管的表达,以研究脑缺血再灌注过程中微血管的损伤与修复。方法 将大鼠随机分为假手术对照组和缺血再灌注(IR)组,两组又各分为再灌注1、3、6、12、24、48、72 h组,动物模型采用全脑缺血模型中的三血管阻塞法建立,用免疫组化法检测脑缺血再灌注不同时间vWF和Laminin的表达规律。结果 假手术各组vWF和Laminin均成强阳性表达,脑缺血再灌注组各时间点表达均低于假手术组,缺血再灌注1 h就出现表达降低,以后逐渐下降,24~48 h表达达最低值,72 h表达又开始上升。结论 大鼠全脑缺血再灌注病理过程中,再灌注1h就出现了脑微血管的损伤,48~72 h出现了血管的修复。
Objective To investigate the expression of von Willebrand factor (vWF) and laminin in brain microvessel after global cerebral ischemia-reperfusion in rats, in order to study the injury and repair of microvascular during cerebral ischemia-reperfusion. Methods The rats were randomly divided into sham-operation control group and ischemia-reperfusion (IR) group. The two groups were divided into reperfusion groups of 1, 3, 6, 12, 24, 48 and 72 h, Three-vessel occlusion was established in the ischemic model. The expression of vWF and Laminin in different time points of cerebral ischemia-reperfusion was detected by immunohistochemistry. Results The expressions of vWF and Laminin in each group were all strongly positive in sham-operated group. The expressions in each group were lower than those in sham operation group at 1 hour after ischemia-reperfusion, and then gradually decreased after 24 h Expression reached the lowest value, 72 h expression began to rise. Conclusion During the process of global cerebral ischemia-reperfusion in rats, cerebral microvascular damage occurred at 1 hour after reperfusion and blood vessel repair occurred 48 to 72 hours after reperfusion.
