目的探讨HBcA g/HBeAg 对慢性乙型肝炎患者BMC中Th1/Th2类细胞应答的影响。方法 用套式PCR法检测64例慢性HBV感染者PBMC中HVB DNA;分别用P H A、HBcAg和HBeAg诱导PBMC体外培养;ELISA法检测PBMC产生Th1类细胞因子(IL-2、IFN-γ)和Th2类细胞因子(IL-4、IL-10)的含量。结果表明HBV DNA阳性组和阴性组相比,无论是在PHA还是在HBcAg/HBeAg诱导下,两 组间 Th1和Th2类细胞因子含量均无显著差异。HBcAg和HBeAg诱导组同PHA诱导组相比,HBcAg诱导下PBMC产生Th1类细胞因子量明显增高;在HBeAg诱导下Th2类细胞因子量明显 增高。结论HBV侵犯PBMC并不影响Th1/Th2应 答;HBcAg和HBeAg可能是影响慢性HBV感染者Th1/Th2类细胞应答的主要因素之一。
Objective To investigate the effect of HBcA g / HBeAg on Th1 / Th2 cell responses in BMC of patients with chronic hepatitis B (HBV). Methods The HVB DNA in PBMC of 64 patients with chronic HBV infection was detected by nested PCR. PBMCs were cultured in vitro with PHA, HBcAg and HBeAg respectively. The levels of Th1 cytokines (IL-2, IFN-γ) and Th2 Cytokines (IL-4, IL-10) content. The results showed that there was no significant difference in the levels of Th1 and Th2 cytokines between the two groups when compared with the negative group, both in PHA and HBcAg / HBeAg-induced groups. HBcAg and HBeAg-induced group compared with PHA induction group, HBcAg-induced PBMC production of Th1-type cytokines increased significantly; HBeAg-induced Th2-type cytokines were significantly higher. Conclusion HBV infection of PBMC does not affect the Th1 / Th2 response. HBcAg and HBeAg may be one of the main factors affecting the Th1 / Th2 response in chronic HBV infection.
