目的:确定骨髓间充质干细胞(BMSCs)在异种胸腺内的演化过程,为进一步进行BMSCs胸腺修饰诱导异种免疫耐受的研究提供可行性依据。方法:取豚鼠BMSCs经分离纯化体外扩增至第3代,并行胎牛血清-Fe2O3和4,6-二咪基-4-联苯基吲哚(DAPI)标记,标记后的豚鼠BMSCs行胸腺注射,7天后进行切片普鲁士蓝-中性红染色和荧光显微镜观察豚鼠BMSCs演变过程。结果:BMSCs移植至异种胸腺中能够生存,并能转化为多种组织细胞成分,即仍具有多向分化能力。结论:异种BMSCs胸腺修饰在诱导异种受体移植免疫耐受方面将是可行的。
OBJECTIVE: To determine the evolution of bone marrow mesenchymal stem cells (BMSCs) in the heterologous thymus, and to provide a feasible basis for the further study of thymus modification induced by BMSCs. Methods: BMSCs of guinea pigs were isolated and purified to the third generation in vitro, and labeled with fetal bovine serum-Fe2O3 and 4,6-diamino-4-biphenylindole (DAPI). The labeled guinea pig BMSCs were cultured in thymus Injected, and stained Prussian blue-neutral red staining and fluorescence microscopy after 7 days to observe the evolution of BMSCs in guinea pigs. Results: BMSCs transplanted into different thymus could survive, and could be transformed into a variety of cell components, that is, they still have multi-directional differentiation ability. CONCLUSION: Thymus modification of xenogenic BMSCs will be feasible in inducing immune tolerance of xenograft recipients.
