采用自行制备的抗IL-5单克隆抗体(McAb)干预哮喘豚鼠模型,以初步了解该单抗在体内的功能。将鸡卵清蛋白致敏成功的豚鼠哮喘模型随机分为两组,分别给予McAb和生理盐水干预,比较致敏前、干预前和干预后外周血嗜酸细胞数(PBEC)变化及两组动物支气管肺泡灌洗液嗜酸细胞差别。结果显示干预后单抗组PBEC较干预前明显降低(P<0.05);单抗组支气管肺泡灌洗液中嗜酸细胞计数较生理盐水组明显降低(P<0.05)。说明所制备的抗IL-5单抗具有在体内中和IL-5的作用。
The anti-IL-5 monoclonal antibody (McAb) prepared by itself was used to interfere the model of asthmatic guinea pigs to understand the function of the McAb in vivo. Egg ovalbumin sensitized guinea pig asthma model were randomly divided into two groups, were given McAb and saline intervention, compared before sensitization, before and after intervention in peripheral blood eosinophil (PBEC) changes and two groups of animals Bronchoalveolar lavage fluid eosinophil differences. The results showed that the PBEC of the monoclonal antibody group was significantly lower than that before intervention (P <0.05). The number of eosinophils in the bronchoalveolar lavage fluid of the monoclonal antibody group was significantly lower than that of the saline group (P <0.05). This shows that the prepared anti-IL-5 mAb has the effect of neutralizing IL-5 in vivo.
