目的 探讨hRARβ基因在白血病发病以及维甲酸类治疗中的细胞生物学作用。 方法 用电穿孔方法将 pcDNA3 hRARβ重组质粒导入缺失hRARβ基因表达的HL 6 0R细胞中 ,并用软琼脂集落形成、细胞形态学、硝基四氮唑蓝 (NBT)还原试验和流式细胞术等方法对其生物学特性进行研究。结果 建立了具有功能性hRARβ基因表达的HL6 0R hRARβ细胞株 ,在 0 .1μmol/LCh5 5作用下其生长抑制率为 5 0 % ,软琼脂培养集落形成率下降 ,细胞分化。结论 在人髓系白血病细胞中恢复外源性hRARβ基因的表达可以恢复该细胞对维甲酸类药物的敏感性 ,从而显著抑制肿瘤细胞生长并诱导其分化成熟
Objective To investigate the cytobiological effects of hRARβ gene in the pathogenesis of leukemia and retinoids treatment. Methods Recombinant pcDNA3 hRARβ plasmid was introduced into HL 60R cells lacking hRARβ gene expression by electroporation, and then used soft agar colony formation, cell morphology, nitroblue tetrazolium (NBT) reduction test, and flow cytometry. Study its biological characteristics. Results The HL6 0R hRARβ cell line with functional hRARβ gene expression was established. The growth inhibition rate was 0.5% under the action of 0.1 μmol/LCh5 5 . The colony formation rate in soft agar decreased and the cells differentiated. Conclusion Restoring the expression of exogenous hRARβ gene in human myeloid leukemia cells can restore the sensitivity of the cells to retinoid drugs, thus significantly inhibiting the growth of tumor cells and inducing their differentiation and maturation.
