目的探讨组织蛋白酶B(CB)在恶性卵巢肿瘤组织中的表达水平及其在肿瘤靶向治疗中的临床意义。方法选取2010年1月至2014年2月在山东省潍坊市人民医院接受手术治疗的72例恶性卵巢肿瘤患者为观察组,同期收治的良性卵巢肿瘤45例患者为对照组,采用蛋白印迹法(western blot)检测患者卵巢组织中CB的表达水平。将A2780传代培养后分为A组和B组,分别加入紫杉醇溶液和紫杉醇溶液+CA-074(Me)溶液,采用四甲基偶氮唑蓝(MTT)技术检测不同时间点的A2780细胞凋亡率。结果对照组CB的表达水平为(0.30±0.07)%,观察组为(0.81±0.14)%,两组比较差异有统计学意义(P<0.05)。0期的CB表达水平为(0.82±0.07)%,I期为(0.83±0.06)%,Ⅱ期为(0.87±0.10)%,Ⅲ期为(0.86±0.09)%,Ⅳ期为(0.88±0.07)%,不同病理分期的患者CB表达水平差异无统计学意义(F=3.12,P>0.05)。A、B两组细胞的凋亡率比较,差异有统计学意义(P<0.05),B组细胞各个时间点的细胞凋亡率均低于A组。各组细胞不同时间点的凋亡率具有统计学差异(P<0.05)。结论 CB不仅参与肿瘤的侵袭与转移,在肿瘤细胞的凋亡中也发挥着重要的作用,在恶性卵巢肿瘤的靶向治疗中具有重要的研究价值。
Objective To investigate the expression of cathepsin B (CB) in malignant ovarian tumor and its clinical significance in tumor targeting therapy. Methods Seventy-two patients with malignant ovarian tumors who underwent surgery in People’s Hospital of Weifang City, Shandong Province from January 2010 to February 2014 were selected as the observation group. Forty-five patients with benign ovarian tumors treated in the same period were selected as the control group by Western blotting western blot) to detect the expression of CB in ovarian tissue. A2780 cells were subcultured and divided into groups A and B, paclitaxel solution and paclitaxel solution + CA-074 (Me) solution respectively, and apoptosis of A2780 cells at different time points was detected by MTT assay rate. Results The expression of CB was (0.30 ± 0.07)% in the control group and (0.81 ± 0.14)% in the observation group, with a significant difference between the two groups (P <0.05). The level of CB in stage 0 was 0.82 ± 0.07%, in stage I was 0.83 ± 0.06%, in stage Ⅱ was 0.87 ± 0.10%, in stage Ⅲ was 0.86 ± 0.09%, in stage Ⅳ was 0.88 ± 0.07% 0.07)%. There was no significant difference in CB expression levels between different pathological stages (F = 3.12, P> 0.05). The apoptosis rates of A and B groups were significantly different (P <0.05). The apoptosis rate of B group was lower than that of A group at each time point. The apoptosis rate of cells in different time points was statistically different (P <0.05). Conclusion CB not only participates in the invasion and metastasis of tumor, but also plays an important role in the apoptosis of tumor cells. CB plays an important role in the targeted therapy of malignant ovarian tumors.
