目的探讨磷脂型DHA(DHA-PC)的抗肿瘤生长作用。方法应用超临界CO2萃取脱除胆固醇技术从鸢乌贼卵中提取DHA-PC。采用MTT法、AO/EB荧光染色法和流式细胞术研究DHA-PC对人肝癌HepG-2细胞增殖、凋亡的影响,并进一步利用小鼠S180移植性肿瘤模型,验证DHA-PC的体内抑瘤效应。结果 DHA-PC能够抑制HepG-2细胞的增殖(P<0.05,P<0.01),诱导细胞凋亡,干扰细胞周期,使细胞停滞在G0/G1期;明显抑制荷瘤小鼠肿瘤的生长,其平均抑瘤率达40.94%。结论 DHA-PC在体内和体外都对肿瘤的生长具有抑制作用。
Objective To investigate the anti-tumor effect of phospholipid DHA (DHA-PC). Methods DHA-PC was extracted from the eggs of Symlectoteuthis oui by supercritical CO2 extraction and removal of cholesterol. The effects of DHA-PC on the proliferation and apoptosis of human hepatocellular carcinoma HepG-2 cells were studied by MTT assay, AO / EB staining and flow cytometry.
