目的:观察温热协同化疗对体外培养的人颊鳞癌BCaCD885细胞的杀伤作用。方法:采用MTT法研究了BCaCD885细胞对43℃温热、几种临床常用化疗药单独应用及联合43℃温热与化疗药时的敏感性。结果:实验表明BCaCD885细胞对温热不敏感,43℃作用40分钟时的细胞死亡率仅7.6%;单独用药时,细胞对ADM最敏感,其次为DDP、VP16、VCR,再次为MMC、BLM,对5Fu、MTX不敏感;43℃温热能显著增强癌细胞对各浓度5Fu、MTX、BLM的敏感性,增强低浓度ADM、DDP、VP16、MMC的敏感性;对VCR无增敏作用。结论:对人颊鳞癌BCaCD885细胞直接杀伤作用很弱的43℃温热对某些化疗药物具有增敏作用,增敏作用的差异与BCaCD885细胞对药物的敏感性、药物浓度、药物作用机制等有关。本实验结果为临床口腔肿瘤的热化疗应用及方案设计提供了理论依据。
Objective: To observe the killing effect of warm synergistic chemotherapy on human cheek squamous cell carcinoma BCaCD885 cells in vitro. METHODS: MTT assay was used to investigate the sensitivity of BCaCD885 cells to 43°C warmth, several commonly used chemotherapy drugs alone and in combination with 43°C warm and chemotherapeutic drugs. Results: The experiment showed that BCaCD885 cells were not sensitive to warming, and the cell death rate at 43°C for 40 minutes was only 7.6%; when used alone, the cells were most sensitive to ADM, followed by DDP, VP16, VCR, and again MMC, BLM, It is insensitive to 5Fu and MTX; 43°C warm energy significantly enhances the sensitivity of cancer cells to 5Fu, MTX, and BLM, and enhances the sensitivity of low concentrations of ADM, DDP, VP16, and MMC; and has no sensitizing effect to VCR. Conclusion: The temperature of 43°C, which has a weak direct killing effect on human buccal squamous cell carcinoma BCaCD885 cells, has a sensitizing effect on certain chemotherapeutic agents. The differences in sensitization effects and the sensitivity of BCACD885 cells to drugs, drug concentration, drug action mechanism, etc. related. The experimental results provide a theoretical basis for the clinical application of thermochemotherapy and design of oral tumors.
