目的在各级脑星形细胞瘤中检测P16、PCNA的表达及DNA含量,探讨这三者能否作为分级及判断预后的指标之一。方法采用免疫组化SP法对136例脑星形细胞瘤行P16、PCNA染色,常规DNA-Feulgen染色。用计算机图象分析仪对以上三项结果分别进行阳性率、阳性指数、DNA指数定量检测,并行q检验分析组间差异。结果在各级星形细胞瘤中P16的表达率为44.5%,平均阳性指数为39.58±16.92,PCNA表达率为95.75%,平均阳性指数为54.7±14.1,瘤细胞核DNA平均指数为1.75±1.69。随着级别增高恶性程度增加,P16表达逐渐降低,而PCNA表达则逐渐增加,DNA指数也逐渐增加。结论提示免疫组化P16、PCNA及细胞核DNA指数可作为脑星形细胞瘤分级及判断预后的指标之一。
Objective To detect the expression of P16 and PCNA and the content of DNA in all levels of brain astrocytoma and to explore whether these three can be used as an index for grading and prognosis. Methods Immunohistochemical SP method for 136 cases of brain astrocytoma P16, PCNA staining, conventional DNA-Feulgen staining. The above three results were respectively analyzed by computer image analyzer for the positive rate, positive index and DNA index, and the q-test was used to analyze the differences between groups. Results The positive rate of P16 was 44.5%, the average positive index was 39.58 ± 16.92, the expression rate of PCNA was 95.75%, the average positive index was 54.7 ± 14.1 and the average nuclear DNA of tumor cells was 1.75 ± 1.69. With the increase of grade, the expression of P16 gradually decreased while the expression of PCNA increased gradually while the DNA index increased gradually. Conclusions Immunohistochemistry P16, PCNA and nuclear DNA index may be used as a marker of brain astrocytoma classification and prognosis.
