目的 探讨肝纤维化形成不同阶段TGFβ1和Ⅰ、Ⅲ型胶原表达的变化规律及其相互关系。方法 建立肝纤维化动物模型;分别用免疫组化法和斑点杂交法检测肝纤维化不同阶段TGFβ1、Ⅰ、Ⅲ型胶原蛋白及其mRNA表达。结果随着肝纤维化程度的加重,TGFβ1、Ⅰ、Ⅲ型胶原蛋白及其mRNA表达量都逐渐增高,各实验组与对照组相比,其差异均有显著性(P<0.05)。结论肝纤维化时,肝内TGFβ1、Ⅰ、Ⅲ型胶原蛋白的表达呈正相关,提示TGFβ1是重要的肝纤维化介质。
Objective To investigate the changes of TGFβ1 and type Ⅰ and type Ⅲ collagen in different stages of hepatic fibrosis and their correlations. Methods The animal model of hepatic fibrosis was established. The expressions of TGFβ1, type Ⅰ and type Ⅲ collagen mRNA and their mRNA in different stages of hepatic fibrosis were detected by immunohistochemistry and dot blot hybridization respectively. Results With the aggravation of hepatic fibrosis, the expression of TGFβ1, type Ⅰ and type Ⅲ collagen and its mRNA were all gradually increased. There was significant difference between each experimental group and control group (P <0.05). Conclusion The expression of TGFβ1, type Ⅰ and type Ⅲ collagen in liver during liver fibrosis is positively correlated, which indicates that TGFβ1 is an important hepatic fibrosis medium.
