目的:研究重组异种骨(RBX)重复植入动物体内引起的二次免疫应答及其对成骨活性的影响。方法:将RBX间隔不同时间重复植入雄性Balb/c小鼠股部肌肉内,通过ELISA测定体液免疫水平,检测局部组织的碱性磷酸酶(ALP)活性反映成骨情况。结果:RBX第1次植入后,血清中可检测到特异性抗体;局部组织中的ALP活性略低于正常小鼠骨骼。间隔3周或6周再次植入RBX,血清中特异性抗体水平明显增高(P<0.05);ALP活性降低(P<0.01)。结论:重复植入RBX可诱发机体产生免疫应答;RBX诱导成骨活性受到抑制,成骨作用延迟。
Objective: To study the secondary immune response induced by repeated implantation of recombinant xenogeneic bone (RBX) into animals and its effect on osteogenic activity. Methods: RBX interspersed at different time intervals were implanted into the muscle of male Balb / c mice. The level of humoral immunity was measured by ELISA. The activity of alkaline phosphatase (ALP) in local tissues was measured to reflect the osteogenesis. Results: After the first implantation of RBX, specific antibodies could be detected in the serum; ALP activity in the local tissues was slightly lower than that in normal mice. RBX was re-implanted 3 weeks or 6 weeks later. The level of serum specific antibody was significantly increased (P <0.05) and ALP activity was decreased (P <0.01). CONCLUSION: Repeated implantation of RBX induces an immune response in the body. RBX-induced osteogenic activity is inhibited and osteogenesis is delayed.
