目的研究金铁锁中寡糖的成分及其结构。方法金铁锁药材采用水提醇沉法经三氯乙酸去蛋白,得到金铁锁粗糖(PTP);PTP依次经DEAE Sepharose F.F.、Sephacryl S-300和Sephadex G25柱层析,最终得到PTP1-1-1;对PTP1-1-1分别用HPLC、GC-MS、甲基化和高碘酸氧化分析进行相关结构研究。结果 HPLC检查PTP1-1-1为单一寡糖;GC-MS糖组成分析和HPLC分子量测定结果推测PTP1-1-1是由1个葡萄糖和6个半乳糖组成的寡糖;甲基化分析、高碘酸氧化分析显示该糖链的连接方式为1,4和1,6连接。结论金铁锁寡糖PTP1-1-1的分离纯化和结构研究丰富了该植物的研究内容。
Aim To study the composition and structure of oligosaccharides in Jindie lock. Methods PTS was obtained by water-alcohol precipitation method and trichloroacetic acid. The PTP was sequentially purified by DEAE Sepharose FF, Sephacryl S-300 and Sephadex G25 column chromatography to obtain PTP1-1-1. PTP1-1-1 were respectively analyzed by HPLC, GC-MS, methylation and periodate oxidation. Results PTP1-1-1 was a single oligosaccharide by HPLC. The results of GC-MS analysis and HPLC analysis suggested that PTP1-1-1 was an oligosaccharide composed of 1 glucose and 6 galactose. Methylation analysis, Periodic acid oxidation analysis showed that the sugar chain was connected by 1, 4 and 1, 6 linkages. Conclusion The isolation, purification and structure study of PTP1-1-1 from Fusarium oxysporum oligosaccharides enriched the research contents of this plant.
