目的:利用荷瘤动物模型初步探讨化学免疫治疗肺癌时不同程序联合应用对肿瘤生长及T细胞功能的影响,以明确最佳治疗方案。方法:建立C57BL/61Leuwis肺癌荷瘤小鼠动物模型,全身应用卡铂及局部应用TNFα、IFNγ,不同程序联合处理荷瘤鼠,观察小鼠肿瘤生长情况。用同位素掺入法检测脾脏T细胞转化能力及脾细胞对IL-2的应答能力。结果:肿瘤生长速率最小者为先卡铂后细胞因子组(P<0.001);其次为卡铂与细胞因子同时组(P<0.01);T细胞转化能力最高组为卡铂、细胞因子同时组(P<0.001)。其他两组升高明显(P<0.05),脾细胞对IL-2的应答能力以同时治疗组和先细胞因子组最高(P<0.05)。结论:不同程序的联合方案对机体T细胞功能影响不同;卡铂与细胞因子同时应用为最佳治疗方案。
OBJECTIVE: To investigate the effects of different procedures combined with chemotherapy on tumor growth and T-cell function in a lung cancer-bearing animal model to identify the best treatment protocol. METHODS: An animal model of C57BL/61 Leuwis tumor bearing mice was established. Systemic application of carboplatin and topical application of TNFα and IFNγ were performed. Tumor-bearing mice were treated with different procedures and tumor growth was observed. Isotope incorporation was used to detect splenic T cell transformation and splenocyte response to IL-2. RESULTS: The smallest tumor growth rate was pre-carboplatin after cytokines group (P<0.001); followed by carboplatin and cytokines simultaneously (P<0.01); the highest T-cell conversion ability was carboplatin, cytokines Simultaneous group (P<0.001). The other two groups were significantly elevated (P<0.05), and the response capacity of spleen cells to IL-2 was highest in the simultaneous treatment group and the first cytokine group (P<0.05). Conclusion: The combination of different procedures has different effects on the function of the body’s T cells; carboplatin and cytokines are the best treatment options at the same time.
