要想长期保存皮肤的活力就必须采用低温或深低温的贮存技术.目前国内外皮库主要使用低温冰箱、液氮(LN_2)等方法来保存皮肤.前者贮存时间较短,一般只能保存一年左右,但贮存条件要求不高,使用较方便,一般单位都能开展.液氮能长期保存皮肤,但贮存条件要求较高,贮存组织需要抗冻处理和慢降温后才能入液氮长期保存.采用慢降温的方法有控制降温(-1℃/分钟)和自然降温两种.一般认为控制降温贮存皮肤的效果最好,细胞冷冻损伤最轻,但要求特殊设备;自然降温不需要特殊设备,只要有干冰、低温冰箱或液氮就能办到,如果使用适当,一样能获得良好效果.干冰降温速率较快,会影响贮存皮肤的活力.近三年我们采用低温冰箱(-80℃)降温贮存皮肤,方法简单、效果良好.假如-80℃以下的冰箱也能承担此项工作,那么液氮贮存皮肤将更为方便.为此我们设计了下列实验,利用冰箱的不同温度降温皮肤,比较了皮肤的降温速率与皮肤活力之间的相互关系.
