目的:建立矮地茶药材中岩白菜素的含量测定方法。方法:矮地茶药材粉碎后过筛,取粉末用甲醇超声提取,未涂渍熔融石英毛细管(75μm×48.5 cm,有效长度40 cm),以25 mmol·L-1硼砂水溶液为缓冲液,运行电压15 k V,温度25℃,内标为肉桂酸,检测波长为240 nm,运行时间10 min。结果:在此条件下岩白菜素与内标及其他成分能达到基线分离,岩白菜素在3.356~167.800μg·ml-1范围内呈良好的线性关系Y=0.034 9X+0.060 6,r=0.997 9,最低检测限为0.839μg·ml-1(S/N=3),最低定量限为3.356μg·ml-1(S/N=10),平均加样回收率为99.6%(RSD=3.0%,n=6),低,中,高浓度日内、日间精密度RSD均小于5%(n=3)。结论:此方法可用于矮地茶药材中岩白菜素的含量测定,具有简便快速、准确可靠、试剂消耗量少的优点。
Objective: To establish a method for determination of bergexin in medicinal materials of dwarf tea. Methods: The powder of Medicago sativa was crushed and sieved. The powder was extracted with methanol by ultrasonic wave. Without fused silica capillary (75μm × 48.5cm, effective length 40 cm), 25 mmol·L-1 borax aqueous solution was used as buffer Voltage 15 k V, temperature 25 ℃, internal standard for cinnamic acid, detection wavelength of 240 nm, running time 10 min. Results: Under these conditions, the concentration of bergenin and the internal standard and other components could be separated from each other. The concentration of bergenin in the range of 3.356-167.800 μg · ml-1 showed a good linearity Y = 0.034 9X + 0.060 6, r = 0.997 9 with the lowest detection limit of 0.839μg · ml-1 (S / N = 3) and the lowest limit of quantification was 3.356μg · ml-1 with an average recovery of 99.6% (RSD = 3.0 %, n = 6). The intra-day and inter-day RSD of low, medium and high concentration were less than 5% (n = 3). Conclusion: This method can be used to determine the content of bergenin in dwarf tea, which has the advantages of simple, rapid, accurate and reliable, less reagent consumption.
