目的:研究女贞子提取物中熊果酸对人肝癌细胞生长抑制作用及对血管内皮生长因子(VEGF),转化生长因子-α(TGF-α)表达的抑制作用,探讨其抗肿瘤机制。方法:女贞子提取物与PLC/PRF/5细胞孵育,以四甲基偶氮唑蓝法(MTT)检测其对人肝癌细胞PLC/PRF/5生长的抑制作用,并通过酶联免疫吸附实验(ELISA)测定细胞培养上清液中VEGF,TGF-α的表达;小鼠右前肢腋皮下接种瘤细胞,连续给药14 d后取瘤组织,检测小鼠肿瘤的抑制率,并以免疫组织化学方法检测瘤组织VEGF,TGF-α表达。结果:女贞子提取物中的熊果酸在体内和体外试验结果均显示抑制人肝癌细胞生长,并对VEGF,TGF-α表达有明显的抑制作用。结论:女贞子提取物具有抗肿瘤作用,抑制VEGF,TGF-α表达可能是其抗肿瘤作用机制之一。
OBJECTIVE: To study the inhibitory effect of ursolic acid on the growth of human hepatocarcinoma cells and the inhibitory effect on the expression of vascular endothelial growth factor (VEGF) and transforming growth factor-α (TGF-α) in Fructus Ligustri Lucidae extracts and to explore its anti-tumor mechanism. Methods: Ligustrum lucidum extract was incubated with PLC / PRF / 5 cells. The inhibitory effect of Ligustrum lucidum extract on PLC / PRF / 5 cells was detected by MTT assay. The expression of VEGF and TGF-α in the cell culture supernatant was measured by ELISA. The tumor cells were inoculated subcutaneously in the right forelimb of the mice. After 14 days of continuous administration, the tumor tissues were taken and the tumor inhibition rate was measured. Histochemical methods were used to detect the expression of VEGF and TGF-α in tumor tissue. Results: Ursolic acid from Fructus Ligustri Lucidi extracts showed inhibitory effects on the growth of human hepatocellular carcinoma cells in vitro and in vivo, and significantly inhibited the expression of VEGF and TGF-α. Conclusion: Ligustrum lucidum extract has anti-tumor effect and inhibition of VEGF and TGF-α expression may be one of its anti-tumor effects.
