我们应用流式细胞术的方法对胆囊肿瘤细胞的p53蛋白进行定量分析,以探讨其与DNA倍体及胆囊肿瘤生物学行为之间的关系,结果报道如下。对象和方法1.对象胆囊肿瘤患者46例,其中胆囊癌36例,腺瘤10例。30例胆囊癌患者获得随访。正常胆囊组织5例作对照。石蜡包埋组织标本。2.DNA检测单细胞悬液样品的制备见文献~[1]。3. p53蛋白检测样品的制备(1)用网搓法制备单细胞悬液。(2)采用间接免疫荧光标记法,其抗体分别为1∶100稀释的鼠抗人p53基因蛋白的单抗
We used flow cytometry to quantitatively analyze p53 protein in gallbladder tumor cells to investigate its relationship with DNA ploidy and gallbladder tumor biological behavior. The results are reported below. Subjects and methods 1. 46 patients with gallbladder tumors, including 36 gallbladder cancers and 10 adenomas. Thirty patients with gallbladder cancer were followed up. Normal gallbladder tissue was used as a control in 5 cases. Paraffin-embedded tissue specimens. 2. DNA detection Single cell suspension samples were prepared as described in [1]. 3. Preparation of samples for detection of p53 protein (1) Single cell suspensions were prepared using a mesh method. (2) Monoclonal antibody against mouse anti-human p53 gene protein diluted 1:100 by indirect immunofluorescence labeling
