针对传统的棉胚珠培养方法因纤维在发育中途夭亡而不能用于研究次生壁增厚和强度形成的机理 ,本文建立了棉胚珠继代培养纤维体系 ,即根据花铃的形态特征选取 类发育状态的胚珠作外植体 ,以 14d为周期连续 3次继代 ,在继代培养基中添加 5 g· L-1活性碳 ,并根据纤维发育不同阶段的特点调整植物生长物质的种类和浓度。结果使纤维重现了植株条件下的发育历程 ,其比强度、长度和细胞壁厚度等品质指标达到或接近植株条件下发育的纤维 ,为研究棉纤维发育和品质形成的机理提供了一个新的实验体系
Aiming at the problem that the traditional method of cotton ovule culture can not be used to study the mechanism of secondary wall thickening and strength formation due to the embryonic development of cotton fiber, the subculture cotton fiber system was established in this paper. State of the ovule as explants to 14d for three consecutive generations of subculture, in the subculture medium added 5 g · L-1 activated carbon, and according to the characteristics of different stages of fiber development to adjust the species and concentration of plant growth substances . As a result, the development of the fiber was reproduced under the condition of the plant, and its specific quality index such as specific strength, length and cell wall thickness reached or was close to that of the plant under development, providing a new experiment for studying the mechanism of cotton fiber development and quality formation system
