目的优选狼牙刺种子中生物碱类成分的提取工艺。方法采用正交试验设计,以提取时间、料液比和提取次数为考察因素,采用高效液相色谱(HPLC)法测定3种生物碱(苦参碱、氧化苦参碱和槐定碱)的含量。色谱柱为Hypersil GOLD C18柱,流动相为乙腈-0.2%磷酸(5:95),流速为1.0 m L·min-1,检测波长为205 nm。结果最佳工艺为10倍量的水回流提取3次,每次3 h。结论该工艺稳定性好,简单可行,可用于狼牙刺中生物碱类成分的提取。
Objective To optimize the extraction process of the alkaloids in the seeds of Spatholobus. Methods The orthogonal design was used to investigate the effects of extraction time, solid-liquid ratio and extraction times on the determination of three alkaloids (matrine, oxymatrine and sophoridine) by high performance liquid chromatography (HPLC) content. The column was Hypersil GOLD C18 with a mobile phase of acetonitrile-0.2% phosphoric acid (5:95) at a flow rate of 1.0 m L · min-1 with a detection wavelength of 205 nm. Results The best process was 10 times the amount of water reflux extraction 3 times, each 3 h. Conclusion The process is stable, simple and feasible, can be used for the extraction of alkaloids from Spike Tooth.
