目的:探讨异氟醚预处理对大鼠缺血/再灌注心肌细胞结构影响的研究。方法:将30只W ister大鼠随机分成3组(每组n=10):假手术组、缺血再灌注损伤组和异氟醚预处理组,建立在体心肌缺血再灌注动物模型。2%异氟醚预处理组缺血再灌注前,吸入异氟醚麻醉1小时,而后处理同缺血再灌注组。实验完毕后,将各组动物心肌组织做组织切片,HE染色,观察各组动物心肌结构变化。结果:假手术组心肌细胞可见部分胞浆溶解;缺血再灌注损伤组心肌细胞胞浆内胞浆溶解呈空泡或网状,细胞界限模糊,可见心肌细胞断裂;2%异氟醚预处理组可见心肌细胞呈短圆柱状,有分支,细胞核呈椭圆形位于中央,胞浆内可见平行排列的肌原纤维,细胞轮廓较清晰,结构较正常。结论:异氟醚预处理对大鼠缺血/再灌注心肌细胞结构具有保护作用。
Objective: To investigate the effect of isoflurane preconditioning on myocardial cell structure in rats during ischemia / reperfusion. Methods: Thirty Wistar rats were randomly divided into 3 groups (n = 10 each): sham-operation group, ischemia-reperfusion injury group and isoflurane preconditioning group. Isoflurane 2% isoflurane pretreatment group before ischemia reperfusion, inhalation isoflurane anesthesia for 1 hour, and then treated with the same group of ischemia-reperfusion. After the experiment, the myocardial tissue of each group was made into tissue sections, HE staining was performed to observe the changes of myocardial structure in each group. Results: Myocardial cells in the sham-operation group were partially cytoplasmolyzed. The cytoplasm of myocardial cells in the ischemic-reperfusion group was vacuolished or reticulated, the cell boundaries were blurred, and the myocardial cells were broken. The isoflurane preconditioning Myocardial cells showed a short cylindrical column, with branches, the nucleus was oval in the central cytoplasm can be seen parallel to the arrangement of myofibrils, cell outline clearer, the structure is normal. CONCLUSION: Isoflurane preconditioning has a protective effect on the myocardial cell structure of ischemia / reperfusion in rats.
