目的 研究甘草酸和苯巴比妥钠对小鼠肝 P45 0 3A酶活性、蛋白含量及 m RNA表达的影响 .方法 给小鼠甘草酸 5 0 mg· kg- 1 、苯巴比妥钠 5 0 mg· kg- 1 或等量注射用水 ig7d后 ,采用利多卡因代谢试验 (MEGX test)、酶联免疫吸咐测定法 (EL ISA)和逆转录聚合酶链反应 (RT- PCR)测定肝微粒体中 P45 0 3A酶活性、蛋白含量及其 m RNA的表达 .结果 与正常对照组相比 ,甘草酸组小鼠尿液 MEGX累积排泄量和肝 P45 0 3A蛋白含量无明显改变 .苯巴比妥钠组小鼠MEGX累积排泄量增加 2 3.4% (P<0 .0 5 ) ,肝 P45 0 3A酶蛋白含量增加 43.4% (P<0 .0 1) . 3组间肝 P45 0 3A酶 m RNA表达无明显差别 .结论 甘草酸对小鼠肝 P45 0 3A酶活性、蛋白含量及其 m RNA表达无明显影响 ;苯巴比妥钠是小鼠肝P45 0 3A酶诱导剂 ,其作用机制位于转录后水平 .
Objective To study the effects of glycyrrhizic acid and phenobarbital on the activity, protein content and m RNA expression of hepatic P45 0 3A in mice. Methods: To mice with glycyrrhizic acid 50 mg·kg-1, phenobarbital sodium 50 The liver microparticles were measured by the MEGX test, EL ISA, and RT-PCR after mg·kg -1 or ig7d equivalent water for injection. Expression of P45 0 3A enzyme activity, protein content and m RNA expression in the body. Results Compared with the normal control group, the cumulative excretion of MEGX and liver P45 0 3A protein content in the urine of the mice in the glycyrrhizic acid group did not change significantly. The cumulative excretion of MEGX increased by 23.4% (P<0.05), and the content of hepatic P45 0 3A enzyme protein increased 43.4% (P<0.01).
