目的观察中药生精汤对雄性不育大鼠睾丸转化生长因子-β1(TGF-β1)表达的影响。方法40只SD大鼠随机分为正常组、模型组、小剂量组、大剂量组,采用腺嘌呤制作雄性大鼠不育模型,小、大剂量组给予生精汤灌胃。造模后30 d处死大鼠,观察睾丸组织形态学变化;采用免疫组织化学法及图像分析系统检测睾丸内TGF-β1的平均光密度;同时测定大鼠体质量和睾丸质量。结果大剂量组与模型组比较睾丸系数明显增加(P<0.05);TGF-β1阳性表达主要见于早期精母细胞、圆形精子细胞以及支持细胞,生精汤大剂量组TGF-β1阳性的表达与模型组比较明显降低(P<0.01)。结论生精汤通过下调TGF-β1的表达实现对睾丸生精功能的调节,对腺嘌呤所致的雄性大鼠不育有一定的治疗效果。
Objective To observe the effect of Shengang Decoction on expression of transforming growth factor-β1 (TGF-β1) in testis of male infertility rats. Methods Forty male Sprague-Dawley rats were randomly divided into normal group, model group, low-dose group and large-dose group. Adenine was used to make the male model of infertility, and the small and high-dose group was given gavage of Shengjing Decoction. Rats were sacrificed 30 days after model establishment. Morphological changes of testis were observed. The average optical density of TGF-β1 in testis was detected by immunohistochemistry and image analysis system. Body weight and testis mass were also measured. Results The testicular coefficient was significantly increased in the high-dose group compared with the model group (P<0.05). The positive expression of TGF-β1 was mainly found in the early spermatocytes, round spermatids and supporting cells. The positive expression of TGF-β1 in the high-dose group of Shengjing Decoction was found. Compared with the model group, it was significantly lower (P<0.01). Conclusion Shengjing Decoction can regulate the spermatogenesis of testis by down-regulating the expression of TGF-β1, and has a certain therapeutic effect on adenine-induced male infertility.
