目的探讨子宫内膜癌组织中基质金属蛋白酶-2(matrix metalloproteinase,MMP-2)及粘附分子CD44拼构变异体6(CD44splice variant 6,CD44v6)的表达,分析其与子宫内膜癌浸润、转移的关系。方法选取73例子宫内膜癌组织标本及同期正常增生期子宫内膜27例为对照组。用免疫组织化学方法检测CD44v6、MMP-2的表达。结果 CD44v6在EC中的阳性表达显著高于正常增生期子宫内膜(P<0.05);CD44v6与肌层浸润深度有关(P<0.05);CD44v6的阳性表达率随淋巴结转移和病理分级增高有下降的趋势,但差异无统计学意义(P>0.05)。MMP-2在子宫内膜癌中的阳性表达显著高于正常增生期子宫内膜(P<0.05);MMP-2仅与肌层有无浸润有关(P<0.05)。CD44v6和MMP-2在EC组织中的表达呈正相关性(rs=0.268,P<0.05)。结论 EC中CD44v6及MMP-2高表达的患者更易发生浸润、转移,且二者有正协同作用。联合检测CD44v6、MMP-2可作为判断子宫内膜癌恶性程度,预测转移和评估预后的生物学指标。
Objective To investigate the expression of matrix metalloproteinase-2 (MMP-2) and adhesion molecule CD44 splice variant 6 (CD44v6) in endometrial carcinoma and its relationship with the invasion, Transfer of the relationship. Methods Totally 73 cases of endometrial cancer tissue samples and 27 cases of normal endometrium in the same period were selected as the control group. Immunohistochemistry was used to detect the expression of CD44v6 and MMP-2. Results The positive expression of CD44v6 in EC was significantly higher than that in normal proliferative endometrium (P <0.05). The expression of CD44v6 was correlated with the depth of myometrial invasion (P <0.05). The positive rate of CD44v6 was decreased with lymph node metastasis and pathological grade However, the difference was not statistically significant (P> 0.05). The positive expression of MMP-2 in endometrial carcinoma was significantly higher than that in normal proliferative endometrium (P <0.05). The expression of MMP-2 was only related to the presence or absence of myometrial invasion (P <0.05). The expression of CD44v6 and MMP-2 in EC tissues was positively correlated (rs = 0.268, P <0.05). Conclusions Patients with high expression of CD44v6 and MMP-2 in EC are more likely to infiltrate and metastasize, and both have positive synergistic effects. Combined detection of CD44v6 and MMP-2 can be used as a biological indicator to predict the malignancy of endometrial carcinoma, predict the metastasis and evaluate the prognosis.
