目的探讨细胞周期G1调控点(checkpoint)相关调节因子细胞周期素D1(CyclinD1)、周期素依赖性蛋白激酶4(CDK4)、多重肿瘤抑制因子(P16MTS1)及视网膜母细胞瘤蛋白(PRb)在HPV6/11相关尖锐湿疣(CA)皮损中的表达和意义。方法采用SP免疫组化染色技术检测27例经PCR检测含HPV6/11DNA的CA皮损和10例正常人皮肤(包皮)中CyclinD1、CDK4、P16MTS1及PRb的表达和分布。结果①正常人皮肤及CA皮损均无CyclinD1表达。②CA皮损基底细胞表达CDK4(均数2.19)较正常人皮肤(均数4.8)降低(P<0.01)。③CA皮损棘细胞表达P16MTS1(均数1.33)较正常人皮肤(均数0)增强(P<0.05)。④CA皮损基底细胞和棘细胞表达PRb(均数各为0.89、0.44)较正常人皮肤(均数均为0)均增强(P<0.05)。结论HPV6/11CA相关皮损的细胞周期G1调控点无明显异常,可能是这类CA皮损不易恶变的原因之一。
Objective To investigate the role of checkpoint-associated cyclin D1, CDK4, P16MTS1 and PRb in HPV6 / 11 related condyloma acuminatum (CA) lesions and its significance. Methods The expression and distribution of CyclinD1, CDK4, P16MTS1 and PRb in 27 cases of CA lesions detected by PCR and 10 cases of normal human skin (foreskin) were detected by SP immunohistochemical staining. Results ① There was no CyclinD1 expression in normal skin and CA lesions. (2) The expression of CDK4 (mean 2.19) in basal cells of CA lesions was lower than that of normal human skin (mean 4.8) (P <0.01). (3) The expression of P16MTS1 (mean 1.33) in cortical lesion cells of CA lesions was significantly higher than that of normal human skin (all 0) (P <0.05). ④CA lesions of basal cells and spine cells expressing PRb (mean, 0.89,0.44) than normal human skin (all 0) increased (P <0.05). Conclusion There is no obvious abnormality in the regulation of cell cycle G1 in HPV6 / 11CA-associated lesions, which may be one of the reasons that lesions of such CA lesions are not easily malignant.
