采用 TCA (三氯醋酸)——冷酒精部分纯化 Namalwa 细胞干扰素,再经 Blue Sepha-rose CL-6B 或 Bluc Dextran Sepharose 4B 柱层析,可将干扰素比活性提高到10~7~10~8IU/mg蛋白,用葡聚糖凝胶 Bio-gel P_(200)和 SDS-PAGE 测得干扰素分子量分别为19,050和18,500。用 DABA-微量荧光法检测了粗制和纯化过程中干扰素制剂中的核酸水平,结果表明 Blue Sepharo-se CL-6B 柱层析可基本廓清最终产物中的 DNA,噬菌体 T_4 DNA 追踪表明,大部分 DNA 在层析中被洗去。
The specific activity of interferon could be increased to 10 ~ 7 ~ 10 ~ 4 by using the purified part of Namalwa cell interferon by TCA - cold alcohol, followed by 4B chromatography on Blue Sepharose CL - 6B or Bluc Dextran Sepharose 4B. 8IU / mg protein, the molecular weight of interferon was 19,050 and 18,500 respectively by Sephadex G-200 and SDS-PAGE. Nucleic acid levels in the interferon preparations during the crude and purification procedures were determined by DABA-microfluorescence. The results showed that the Blue Sepharo-se CL-6B column chromatography could basically clear the DNA in the final product. The phage T 4 DNA tracing showed that the large Part of the DNA was washed away in the chromatography.
