探明色氨酸在壳角蛋白中的存在与含量。方法:实验设计用荧光法、氨基酸自动分析仪和对二甲氨基苯甲醛(DAB)化学分析法三种方法定性定量对比分析检测。结果:三种检测方法的分析结果均显示壳角蛋白内有色氨酸存在,占壳角蛋白的百分含量分别为:荧光法:2.50%~2.58%,氨基酸自动分析仪:1.95%~2.27%,DAB化学分析法:2.51%~2.64%。结论:存在于珍珠及其贝壳内的壳角蛋白的氨基酸组份中,有相对含量较多的色氨酸,这在某种程度上可以解释珍珠具有安神镇定药效的机理。
To determine the presence and content of tryptophan in shell keratin. Methods: The experimental design was qualitatively and quantitatively analyzed by fluorescence method, amino acid automatic analyzer and dimethylaminobenzaldehyde (DAB) chemical analysis method. Results: The results of the three assays showed that tryptophan was present in the shell keratin. The percentage of keratin contained in the shell keratin was: Fluorescence: 2.50% to 2.58%, Amino Acid Analyzer: 1 .95% to 2.27%, DAB Chemical Analysis: 2.51% to 2.64%. Conclusion: There is a relatively high content of tryptophan in the amino acid component of shell keratin present in pearls and shells, which can explain the mechanism of calming down the efficacy of pearl in some extent.
