目的探讨毛菊苣种子药材的鉴别方法,完善毛菊苣种子的药材质量标准。方法采用薄层色谱法,对毛菊苣种子中活性成分秦皮乙素进行鉴别;以高效液相色谱法测定秦皮乙素含量。选用Phenomenex Luna C18柱(250 mm×4.6 mm,5μm),流动相为甲醇-0.3%甲酸(25∶75),流速1.0 ml.min-1,检测波长349 nm,进样量10μl。结果薄层色谱中秦皮乙素斑点清晰,无干扰。高效液相色谱法精密度、重复性良好;秦皮乙素在0~0.2μg范围内有较好的线性关系,3批药材中秦皮乙素含量分别为0.099 7,0.110 9,0.097 2 mg.g-1。结论该方法稳定、可靠,为评价毛菊苣种子药材的质量提供科学依据。
Objective To explore the identification method of Mao chicory seeds medicinal materials and improve the quality standards of medicinal materials of Mao chicory seeds. Methods The content of aesculetin was identified by TLC. The content of aesculetin was determined by high performance liquid chromatography (HPLC). The Phenomenex Luna C18 column (250 mm × 4.6 mm, 5 μm) was used. The mobile phase consisted of methanol-0.3% formic acid (25:75) and the flow rate was 1.0 ml.min-1. Results Aesculin spots in TLC were clear and non-interfering. The precision and repeatability of HPLC were good. There was a good linear relationship between the content of aesculetin and 0 ~ 0.2μg. The contents of aesculetin in three batches were 0.099 7,0.110 9,0.097 2 mg.g respectively. -1.
