目的:建立复方甘草片中吗啡与磷酸可待因的含量测定方法。方法:液相色谱电喷雾串联质谱仪,采集方式MRM(多反应监测),色谱柱Zorbax Eclipse SB C18(2.1 mm×100 mm,3.5μm),流动相:乙腈~(-1)5 mmol·L~(-1)乙酸铵,梯度洗脱。结果:吗啡与可待因的母离子质核比分别为286.0、300.0,定量离子为165.0、165.0,吗啡在25~500 ng·ml~(-1)范围内线性良好,r=0.999 5,平均回收率为103.1%,磷酸可待因在0.75~150 ng·ml~(-1)范围内线性良好,r=0.999 5,平均回收率为101.5%,RSD=1.7%(n=6)。结论:本方法灵敏度高,前处理简单,可用于复方甘草片的质量控制。
Objective: To establish a method for the determination of morphine and codeine phosphate in compound licorice tablets. Methods: The liquid chromatography-electrospray tandem mass spectrometry (MS-MS / MS) was used to collect MRM (multi-reaction monitoring) and the column was Zorbax Eclipse SB C18 (2.1 mm × 100 mm, ~ (-1) ammonium acetate, gradient elution. RESULTS: The ratios of proton to proton in morphine and codeine were 286.0 and 300.0, respectively, and the quantitation ions were 165.0 and 165.0. Morphine showed good linearity in the range of 25-500 ng · ml -1, r = 0.999 5. The mean The recovery was 103.1%. Codeine phosphate had a good linearity in the range of 0.75-150 ng · ml -1, r = 0.999 5, with an average recovery of 101.5% and RSD = 1.7% (n = 6). Conclusion: The method has high sensitivity, simple pretreatment and can be used for quality control of compound liquorice tablets.
