目的探讨模拟失重大鼠心室肌自噬相关基因Beclin-1和微管相关蛋白轻链3(LC3)的表达变化。方法雄性SD大鼠12只,随机分为对照组和模拟失重组。模拟失重4周后取心室肌组织,采用RT-PCR和Western Blot方法测定Beclin-1和LC3 mRNA及蛋白表达的变化。结果与对照组相比,模拟失重4周后大鼠心室肌Beclin-1和LC3 mRNA及蛋白表达均显著增强(P<0.05)。结论模拟失重可诱导大鼠心室肌自噬相关基因Beclin-1和LC3的高表达,提示模拟失重可能引起了心室肌自噬的激活。
Objective To investigate the expression changes of autophagy-related genes Beclin-1 and microtubule associated light chain 3 (LC3) in simulated weightless rats. Methods Twelve male SD rats were randomly divided into control group and simulated weightlessness group. After 4 weeks of simulated weightlessness, ventricular myocytes were taken and the mRNA and protein expressions of Beclin-1 and LC3 were determined by RT-PCR and Western Blot. Results Compared with the control group, mRNA and protein expression of Beclin-1 and LC3 in the ventricular myocardium were significantly increased after 4 weeks of simulated weight loss (P <0.05). Conclusion Simulated weightlessness can induce high expression of autophagy-related genes Beclin-1 and LC3 in rats, suggesting that simulated weightlessness may induce the activation of autophagy in ventricular myocardium.
