目的 探讨树舌多糖GF对小鼠HepA瘤细胞Rb基因表达的影响。方法 本实验用链霉菌抗生素蛋白 -过氧化酶免疫组化法来测定细胞中Rb蛋白含量 ,通过多媒体图像分析系统分析实验结果。结果 树舌多糖GF组、猪苓多糖组与荷瘤对照组比较 ,差异均非常显著 (P <0 .0 1) ,即树舌多糖GF、猪苓多糖能促使HepA瘤细胞中Rb基因表达明显增强 ;树舌多糖组与猪苓多糖组比较 ,差异非常显著 (P <0 .0 1) ,即树舌多糖GF更能明显增强Rb基因表达 ,优于猪苓多糖。结论 树舌多糖GF抗瘤作用机理之一是作用于抑癌基因Rb并使之表达增强。
Objective To investigate the effect of polysaccharide GF on the expression of Rb gene in mouse HepA tumor cells. Methods In this experiment, Streptococcus aureus protein-peroxidase immunohistochemical method was used to determine the content of Rb protein in cells. The experimental results were analyzed by multimedia image analysis system. Results The results showed that the differences of the polysaccharides from the GF group and the polyporus umbellatus polysaccharide group were significantly different from the tumor-bearing control group (P < 0.01). The results showed that the polysaccharides GF and Polyporus umbellatus polysaccharides could promote the expression of Rb gene in HepA tumor cells. There was a significant difference between the polysaccharide group and the Polyporus umbellatus polysaccharide group (P < 0.01). That is to say, Ganoderma lucidum polysaccharide GF can obviously enhance the expression of Rb gene, which is superior to Polyporus umbellatus polysaccharide. Conclusion One of the mechanisms of the anti-tumor effect of polysaccharide GF on the tree tongue is to act on the tumor suppressor gene Rb and increase its expression.
