目的检测nucleostemin(NS)基因在人乳腺肿瘤组织中的表达情况,探讨NS基因与乳腺肿瘤恶性程度的关系。方法提取组织总RNA,用半定量RT-PCR方法检测NS基因在乳腺肿瘤中的表达情况,分析NS基因与乳腺肿瘤病理分级、组织学类型和转移的关系。结果乳腺肿瘤组织中NS基因有不同程的表达,病理分级越高,NS基因的表达量越高(F=41.776,P=0.000),正常乳腺组织中没有NS基因的表达;转移组NS基因表达量高于无转移组(t=6.033,P=0.000),NS基因的表达量高低与乳腺肿瘤组织学类型无关(F=2.163,P=0.48)。结论乳腺肿瘤组织中有NS基因表达,NS基因的表达量高低与乳腺肿瘤病理分级和转移有关,与组织学类型无关。
Objective To detect the expression of nucleostemin (NS) gene in human breast neoplasm and to explore the relationship between the NS gene and the malignancy of breast neoplasm. Methods Tissue total RNA was extracted and the NS gene expression in breast cancer was detected by semi-quantitative RT-PCR. The relationship between NS gene and pathological grade, histological type and metastasis of breast cancer was analyzed. Results The NS gene expression in different stages of breast cancer was different. The higher the pathological grade was, the higher the NS gene expression was (F = 41.776, P = 0.000). The NS gene expression was not found in normal breast tissue. The level of NS gene was higher in non-metastatic group than in non-metastatic group (t = 6.033, P = 0.000). The expression level of NS gene was not correlated with histological type of breast tumor (F = 2.163, P = 0.48). Conclusion The expression of NS gene in breast cancer tissue is related with the expression of NS gene in breast cancer, which is not related to the histological type.
