目的正交设计优选理气散结颗粒的提取工艺。方法采用HPLC测定芍药苷含量,色谱柱为Diamansil C18柱,流动相为乙腈(A)-0.1%磷酸溶液(B)梯度洗脱(0~14min,0%~15%B;14~15min,15%~90%B;15~25min,90%B;25~26min,15%~90%B;26~30min,85%B),流速为1.0 ml·min-1;柱温为30℃;检测波长λ=230 nm。以芍药苷提取量和干膏率的综合评分为指标,在单因素试验的基础上,通过正交试验考察提取时间、料液比、提取次数对提取工艺的影响。结果最佳提取工艺为加10倍量水提取两次,每次1h;芍药苷提取量为28.96 mg·g-1,干膏得率34.07%。结论通过正交设计优选的提取工艺简单、经济、合理、可行,为医院制剂-理气散结颗粒的应用提供参考。
Objective To optimize the orthogonal design of Qi San San particles extraction process. Methods HPLC was used to determine the content of paeoniflorin on a Diamansil C18 column. The mobile phase consisted of a gradient of acetonitrile (A) - 0.1% phosphoric acid (B) (0-14 min, 0-15% B; % ~ 90% B; 15 ~ 25min, 90% B; 25 ~ 26min, 15% ~ 90% B; 26 ~ 30min, 85% B) at a flow rate of 1.0 ml · min- Wavelength λ = 230 nm. Based on the single factor test, taking the comprehensive score of the extraction amount of paeoniflorin and the dry paste as the index, the influence of extraction time, the ratio of solid to liquid and the number of extraction on the extraction process was investigated by orthogonal test. Results The optimum extraction process was extraction of 10 times the amount of water twice, each time 1h; extraction of paeoniflorin was 28.96 mg · g-1, the dry paste yield was 34.07%. Conclusion The optimal extraction process by orthogonal design is simple, economical, reasonable and feasible, which can provide a reference for the application of hospital preparation - Liqi Sanjie Granules.
