目的:研究肝纤维化过程中胶原成分的变化规律.方法:制作四氯化碳肝纤维化模型,测定肝组织羟脯氨酸含量,用VG、RF染色显示Ⅰ、Ⅲ型胶原并进行计算机图像分析,放免法测定肝匀浆及血清Ⅲ型前胶原(PCⅢ)水平,应用上述指标反映肝纤维化过程中胶原的变化规律.结果:随实验时间延长,肝内胶原含量逐渐增加,肝纤维化早、中期以Ⅲ型胶原增生为主,肝纤维化后期及肝硬化期以Ⅰ期胶原增生为主.血清PCⅢ与肝内胶原变化基本同步.结论:肝内胶原增加是肝纤维化过程的病理基础,血清PCⅢ可作为反映肝纤维的指标.
OBJECTIVE: To study the changes of collagen composition during liver fibrosis.Methods: CCl4-induced hepatic fibrosis model was established, and the content of hydroxyproline in liver tissue was determined. Type I and type III collagen were visualized by VG and RF, The level of collagen Ⅲ in liver homogenate and serum type Ⅲ procollagen (PCⅢ) was measured by radioimmunoassay, and the changes of collagen in liver fibrosis were observed by using the above indexes.Results: With the prolongation of experimental time, the content of intrahepatic collagen gradually increased and liver fibrosis The main type Ⅲ collagen hyperplasia in the early and middle stages, the stage Ⅰ collagen hyperplasia in the late liver fibrosis stage and the liver cirrhosis stage.The serum PCⅢ basically agrees with the intrahepatic collagen change.Conclusion: Intrahepatic collagen increase is the pathological process of hepatic fibrosis Basis, serum PC Ⅲ can be used as an indicator of liver fiber.
