目的:研究白细胞介素6(IL-6)在子宫内膜异位症(endometriosis,EM)发病中的作用。方法:采用酶联免疫吸附试验(ELISA)检测体外培养EM患者(12例)的异位内膜细胞和在位内膜细胞培养上清液中IL-6水平并与10例正常子宫内膜细胞作对照;以及肿瘤坏死因子α(TNF-α)对异位内膜细胞分泌IL-6的影响。结果:三种内膜细胞培养上清液中IL-6水平明显不同。异位内膜细胞分泌的IL-6显著高于相应的在位内膜(P<0.01),而后者又明显高于正常子宫内膜(P<0.05),即IL-6分泌量异位内膜细胞>在位内膜细胞>正常子宫内膜细胞。TNF-α作用24h后可明显促进异位内膜细胞IL-6的分泌(P<0.01)。结论:异位内膜细胞可自主分泌高水平IL-6,TNF-α进一步诱导其分泌,与EM患者腹腔免疫和炎症反应相关。EM患者在位内膜细胞分泌IL-6的活性明显升高,可能导致其异位种植。
Objective: To investigate the role of interleukin-6 (IL-6) in the pathogenesis of endometriosis (EM). Methods: The levels of IL-6 in ectopic endometrial cells and eutopic endometrial cell culture supernatants of EM patients (12 cases) cultured in vitro were detected by enzyme linked immunosorbent assay (ELISA) and compared with 10 cases of normal endometrial cells As control; and the effect of tumor necrosis factor alpha (TNF-α) on secretion of IL-6 by ectopic endometrial cells. Results: The level of IL-6 in the three endometrial cell culture supernatants was significantly different. Ectopic endometrial cells secrete IL-6 was significantly higher than the corresponding eutopic endometrium (P <0.01), which was significantly higher than the normal endometrium (P <0.05), that is, the secretion of IL-6 in the ectopic Membrane cells> Eutopic endometrial cells> Normal endometrial cells. The effect of TNF-α for 24h on secretion of IL-6 in ectopic endometrial cells (P <0.01). CONCLUSION: Ectopic endometrial cells can secrete high levels of IL-6 and induce TNF-α secretion, which is associated with peritoneal immune response and inflammatory reaction in patients with EM. EM patients in endometrial cells secrete IL-6 activity was significantly increased, may lead to its ectopic implantation.
