目的探讨Ⅰ型变态反应、Th1、Th2类细胞因子在小鼠分泌性中耳炎中耳黏膜的表达情况。方法采用卵清白蛋白致敏并耳内激发小白鼠制成分泌性中耳炎(OME)模型。以免疫组织化学方法检测实验组、对照组中耳黏膜IL4、IL5、IFNγ、IL12的表达情况,同时作病理检查,了解嗜酸粒细胞浸润与各细胞因子的关系。结果实验组Th2类细胞因子IL4、IL5表达明显增加,伴相应的中耳黏膜嗜酸粒细胞浸润,而Th1类细胞因子IFNγ、IL12明显减少,且嗜酸粒细胞浸润与IL5呈正相关(γ=0.951,P<0.05)。结论Ⅰ型变态反应是OME发病的重要原因,而Th2类细胞因子在其发病过程中起着重要的作用。
Objective To investigate the expression of type Ⅰ allergic reaction, Th1 and Th2 cytokines in middle ear mucosa of secretory otitis media in mice. Methods The secretory otitis media (OME) model was induced by ovalbumin sensitization and ear stimulation in mice. Immunohistochemistry was used to detect the expression of IL4, IL5, IFNγ and IL12 in the middle ear mucosa of the experimental group and the control group. At the same time, pathological examination was performed to find out the relationship between eosinophil infiltration and various cytokines. Results The expression of Th2 cytokines IL4 and IL5 in the experimental group was significantly increased with the corresponding eosinophilic infiltration of the middle ear mucosa, while the levels of Th1 cytokines IFNγ and IL12 were significantly decreased and eosinophil infiltration was positively correlated with IL5 (γ = 0.951, P <0.05). Conclusion Type Ⅰ allergic reaction is an important cause of OME, and Th2 cytokines plays an important role in the pathogenesis of OME.
