目的 分离日本血吸虫可溶性成虫抗原 (AWA)中的组分抗原 ,分析其诱导的抗血吸虫的免疫保护性。方法 电泳层析法分离日本血吸虫AWA中的组分抗原 ,计算各组分抗原的分子量 ,并分析其免疫学活性。将各组分抗原分别免疫小鼠 ,攻击感染日本血吸虫尾蚴 ,感染后 4 5d剖杀 ,观察各组分抗原的免疫保护效果。结果 采用电泳层析法分离获得多个组分抗原 ,选择收集其中 4个较纯的组分抗原 ,分子量分别为 5 2kD、6 3kD、6 7kD和 74kD ;其中 6 3kD和 6 7kD抗原与感染血清有强的反应 ,而 5 2kD和 74kD抗原则反应弱 ;4种组分抗原均与AWA的免疫血清反应。 6 3kD抗原诱导的免疫保护力水平较5 2kD、6 7kD和 74kD抗原好。结论 电泳层析法分离的 4个血吸虫组分抗原有良好的免疫原性 ,且 6 3kD抗原有一定的免疫保护性。
Objective To isolate the component antigens of soluble adult worm antigen (AWA) of Schistosoma japonicum and analyze its immunological protection induced by anti-schistosomes. Methods The antigen of antigen of Schistosoma japonicum AWA was isolated by electrophoresis, the molecular weight of each antigen was calculated, and its immunological activity was analyzed. The antigens of each component were respectively immunized with the mouse and challenged with cercariae of Schistosoma japonicum. The infected mice were killed at 45 days after infection and the immunoprotective effect of each component antigen was observed. Results A number of antigens were isolated by electrophoresis. Four of the pure antigens were selected and their molecular weights were 52kD, 63kD, 67kD and 74kD, respectively. Among them, There was a strong response, while the 5 2kD and 74kD antigens reacted weakly; all four antigens reacted with AWA immune sera. 6 3kD antigen-induced immune protection than 5 2kD, 67kD and 74kD antigens better. Conclusion The four antigens of Schistosoma japonicum isolated by electrophoresis showed good immunogenicity, and the 6 3 kD antigens had some protective immunity.
