目的:了解慢性粒细胞白血病(CML)患者杀伤细胞免疫球蛋白样受体(killer cell immunoglobulin-like receptor,KIR)基因型及单倍型,为探讨KIR在慢性粒细胞白血病中发挥的作用奠定基础。方法:采用PCR-SSP方法对84例慢性粒细胞白血病患者进行KIR基因分型。结果:框架基因2DL4、3DL2、3DL3及3DP1存在于所有个体中,其他较为高频的基因依次为2DL1、2DL3、2DP1及3DL1,表型频率均>80%。CML患者的KIR2DL2和KIR2DS2表型频率较健康人有所降低,但除2DL5B(OR=0.208,P<0.05)外,分析的所有基因在CML患者和健康人间的分布均不具有显著差异。结论:基因KIR2DL5B可能与CML之间存在相关性。
OBJECTIVE: To understand the killer cell immunoglobulin-like receptor (KIR) genotype and haplotype in patients with chronic myeloid leukemia (CML) and lay a foundation for exploring the role of KIR in chronic myeloid leukemia . Methods: KIR genotyping was performed in 84 patients with chronic myelogenous leukemia using PCR-SSP method. RESULTS: The framework genes 2DL4, 3DL2, 3DL3 and 3DP1 were present in all individuals. Other higher-frequency genes were 2DL1, 2DL3, 2DP1 and 3DL1 with the phenotypic frequencies> 80%. The frequencies of KIR2DL2 and KIR2DS2 phenotypes in CML patients were lower than those in healthy subjects, but the distribution of all the genes analyzed was not significantly different between CML patients and healthy subjects except 2DL5B (OR = 0.208, P <0.05). Conclusion: There is a correlation between the gene KIR2DL5B and CML.
