取3529×Oro(F_1)及其亲本的新鲜叶片,经组织捣碎、离心、得线粒体;用SDS破线粒体,过氯酸钠、氯仿除蛋白。透析纯化后,用核糖核酸酶除线粒体R NAS;再经Sepharose4 B柱层析得纯净的线粒体DNA(mitDNA)。电镜观察制备的mitDNA为共价闭合环状分子。3529、3529×Oro(F_1)、Oro的mitDNA的Tm值分别为85.3℃、85.0℃、86.0℃;增色值分别为23%、29%、5%;(G+C)%含量分别为39、38、41。杂种mitDNA与亲本的差异或接近也反映在其性状1上。如3529、杂种(F_1)、Oro的株高(cm)分别为163.1、173.2、411.8;全株有效果数分别为323.7、407.0、340.5;每果籽粒数分别为13.1、14.6、11.4;千粒重(g)分别为5.20、4.47、3.60;含油量(%)分别为38.75、42.60、37.10;单株产量(g)分别为14.2、18.7、12.4;亩产量(kg)分别为87、157、76,都优于双亲而更接近于母本3529。
