目的 建立简便易行的单纯疱疹病毒I型 (HSV_1)潜伏感染动物模型。方法 BALB c小鼠 6 0只随机分为 3组 ,分别为鼻黏膜病毒滴入组、尾静脉接种组和角膜划痕接种组 ,进行单纯疱疹病毒I型接种。 6周后行小鼠三叉神经节病毒抗原检测和病毒DNA片段检测 ,以确认潜伏感染的存在。结果 尾静脉接种组小鼠三叉神经节病毒DNA片段的检出率为 9 12 ,角膜划痕组小鼠三叉神经节病毒DNA片段的检出率为 13 16 ,均高于鼻黏膜接种 12 2 0 ;小鼠三叉神经节病毒抗原表达均为阴性 ;尾静脉接种组死亡率 4 15 ,高于其他两组。结论 鼻黏膜病毒滴入、尾静脉注射和角膜划痕接种均可建立小鼠HSV_1潜伏感染模型 ,以角膜划痕法引起疱疹病毒急性感染症状轻恢复快 ,潜伏感染发生率高 ,为最理想的建立HSV_1潜伏感染的方法。
Objective To establish an animal model of latent infection with herpes simplex virus type I (HSV_1). Methods Sixty BALB / c mice were randomly divided into three groups: nasal mucosal virus instillation group, tail vein inoculation group and corneal scratching inoculation group, respectively, for herpes simplex virus type I inoculation. Trigeminal ganglia virus antigen detection and viral DNA fragment detection were performed 6 weeks later to confirm the existence of latent infection. Results The detection rate of the DNA fragment of the trigeminal ganglion virus in the tail vein inoculation group was 9 12. The detection rate of the DNA fragment of the trigeminal ganglion virus in the corneal scratch group was 13 16, which was higher than that of the nasal mucosa ; The expression of antigen of mouse trigeminal ganglion virus was negative; the mortality rate of tail vein inoculation group was 4 15, higher than the other two groups. Conclusions Nasal mucosal virus instillation, tail vein injection and corneal scar vaccination can establish a latent infection model of HSV-1 in mice, and the corneal scratching method can cause acute recovery of herpesvirus with rapid recovery of symptoms and high incidence of latent infection, which is the most ideal Establish a method of latent infection with HSV_1.
