为了建立同时测定苜蓿中4种黄酮的高效液相色谱(HPLC)法,并比较它们的含量,试验采用Eclipse XDB-C_(18)色谱柱(150 mm×4.6 mm),以乙腈(A)-0.5%的磷酸溶液(B)为流动相,梯度洗脱,流速为0.8 m L/min,检测波长为254 nm,柱温为25℃。结果表明:木犀草素在苜蓿花中含量达到27.26μg/g,染料木素和芒柄花黄素在苜蓿花中的含量也均较高;苜蓿根中的黄酮素含量较低,尤其是槲皮素在苜蓿根中含量极其微少。
In order to establish a method for simultaneous determination of four flavonoids in alfalfa by HPLC and to compare their contents, the determination was performed on an Eclipse XDB-C 18 column (150 mm × 4.6 mm) 0.5% phosphoric acid solution (B) as mobile phase, gradient elution, flow rate 0.8 m L / min, detection wavelength 254 nm, column temperature 25 ℃.
